چکیده

Phoslactomycins (PLMs) A–F, produced by actinomycetes are polyketide-type antibiotics derived from a hydroxycyclohexanecarboxylic acid or a cyclohexanecarboxylic acid starter unit. Feeding experiments with [2-C]shikimic acid indicated that the C-18 carbon of PLMs comes from C-5 of shikimate. Further feeding studies of cis and trans-3-hydroxy[7-C]cyclohexanecarboxylic acid, [7-C]and [H11]cyclohexanecarboxylic acid have suggested that the starter unit in the PLM biosynthesis is not cis-3hydroxycyclohexanecarboxylate but cyclohexanecarboxylate and that PLM-B is produced initially, and subsequently converted to other analogs by hydroxylation and acylation. q 2003 Elsevier Ltd. Enhancement and selective production of phoslactomycin B, a protein phosphatase IIa inhibitor, through identification and engineering of the corresponding biosynthetic gene cluster.

Phoslactomycins (PLMs), potent and selective inhibitors of serine threonine phosphatases, are of interest for their antitumor and antiviral activity. Multiple analogs and low titers in the fermentation process have hampered the development of this class of natural products. The entire 75-kb PLM biosynthetic gene cluster of Streptomyces sp. HK-803 was cloned, sequenced, and analyzed. The loading...

In vivo and in vitro effects of thiolactomycin on fatty acid biosynthesis in Streptomyces collinus.

A stable-isotope assay was used to analyze the effectiveness of various perdeuterated short-chain acyl coenzyme A (acyl-CoA) compounds as starter units for straight- and branched-chain fatty acid biosynthesis in cell extracts of Streptomyces collinus. In these extracts perdeuterated isobutyryl-CoA was converted to isopalmitate (a branched-chain fatty acid), while butyryl-CoA was converted to pa...

Modular polyketide synthases and cis double bond formation: establishment of activated cis-3-cyclohexylpropenoic acid as the diketide intermediate in phoslactomycin biosynthesis.

Phoslactomycins (PLMs), exemplified by PLM B (Figure 1), are a unique class of antitumor, antiviral, and antifungal polyketide natural products.1,2 The antitumor activity of PLMs is attributed to a potent and selective inhibition of protein Ser/Thr phosphatase 2A (PP2A).3 The PLM biosynthetic gene cluster from Streptomyces sp. HK803 has been cloned and sequenced.4 The PLM polyketide synthase (P...

The Streptomyces peucetius dpsC gene determines the choice of starter unit in biosynthesis of the daunorubicin polyketide.

The starter unit used in the biosynthesis of daunorubicin is propionyl coenzyme A (CoA) rather than acetyl-CoA, which is used in the production of most of the bacterial aromatic polyketides studied to date. In the daunorubicin biosynthesis gene cluster of Streptomyces peucetius, directly downstream of the genes encoding the beta-ketoacyl:acyl carrier protein synthase subunits, are two genes, dp...

Mutational biosynthesis of a FK506 analogue containing a non-natural starter unit.

A FK506 analogue containing a non-natural starter unit was obtained through mutasynthesis by feeding cultures of Streptomyces sp. KCTC 11604BP fkbO deletion mutant with 3-cyclohexene-1-carboxylic acid. The structure of the new compound, 32-dehydroxy-FK506, and its biological activities were determined.
